日本・タイ感染症共同研究センター  ウイルス感染部門

ウイルス感染部門では、タイおよびわが国を含むアジア諸国で感染が繰り返されているウイルス性腸管感染症と蚊媒介性感染症を研究対象に、タイ王国保健省医科学局の研究者とともに研究を推進しています。

 

熱帯に位置するタイ王国には、様々な蚊媒介性ウイルス感染症が蔓延しています。それら蚊媒介性ウイルスは、タイ王国と国交が盛んな日本を含めた世界各国へ伝播する可能性があります。そのため、蚊媒介性ウイルスの基礎的研究に基づいた感染への対策法の確立は、防疫における重要な課題です。タイ王国に分布が認められる蚊媒介性ウイルスのうち、デング熱や重症型デング出血熱の原因因子であるデングウイルスの抗体依存性感染増強(Antibody Dependent Enhancement of Infection: ADE)に焦点を当て研究活動を行っています。迅速ADE試験法の開発、デング熱や重症型デング出血熱の重症化予測を目標としたコホート研究、それら疾患の重症化に関わる機序の解明を目指したADEに関与する因子の解析を行っています。
 

例年、世界各国に認められるノロウイルスの感染を原因とする急性胃腸炎は、公衆衛生における問題の一つです。ウイルスゲノムの変異や組換えによるウイルスの多様化が、宿主免疫からの回避と持続的な蔓延を可能にしていると考えられています。しかしながら、どの様にウイルスが多様化するのか、どの様にウイルスがヒト社会で保持されているのかは十分に理解されていません。疫学調査に基づいたゲノム変異を伴う流行株の変化の把握とノロウイルスの生活環の解明を目指して研究を行っています。 

  • ADE試験(従来法)の実験行程前半は抗体とウイルスと細胞を混ぜて培養するのみの単純な作業であるが、後半で感染細胞を特異抗体で染色したり、感染細胞を顕微鏡下でカウントしたりするのに相当な時間と労力を必要とした。

  • そこで、抗原にSRIP(Single Round Infectious Particle)と呼ばれる1回感染性粒子を用いた迅速ADE試験法を開発した。本法によって、ADE試験の特に後半の実験行程が簡便となり、さらに短時間で結果を得られるようになった。
