Abstract: The development of protective immunity against malaria is a slow and poorly understood process that has a significant strain-specific component to it. This means that a specific malaria strain is able to induce a protective immune response against itself, but less so against genetically unrelated strains. In order for strain-specific protective immunity to exist, there must be genetic polymorphism within the antigenic targets of this immunity. We are investigating which parasite protein(s) confers strain-specific protective immunity against the rodent malaria parasite Plasmodium chabaudi chabaudi. Using a combination of genetic techniques we have identified a region of the P.c.chabaudi genome that confers strain-specific protective immunity and which contains the gene encoding the Merozoite Surface Protein 1. This same chromosomal region also confers strain-specific protective immunity in other genetically distinct strains of rodent malaria parasite. Our evidence indicates that a single genetic locus may be sufficient to control this very important component of naturally acquired immunity against malaria.
